Enrichment of Skeletal Stem Cells from Human Bone Marrow Using Spherical Nucleic Acids

Human bone marrow (BM)-derived stromal cells contain a population of skeletal stem (SSCs), with the capacity to differentiate along osteogenic, adipogenic, and chondrogenic lineages, enabling their application clinical therapies. However, current methods isolate enrich SSCs from human tissues remain, at best, challenging in absence specific SSC marker. Unfortunately, none proposed markers alone can homogeneous cell ability form bone, cartilage, adipose tissue humans. Here, we have designed DNA-gold nanoparticles able identify sort displaying mRNA signatures. The approach demonstrates significant enrichment attained isolation SSCs, potential therein enhance our understanding biology translational applications.
